dPCR LNA Mutation Assays เป็นชุด kit ที่สามารถตรวจวัดการกลายพันธุ์ซึ่งอ้างอิงจากฐานข้อมูล COSMIC ซึ่ง Kit นี้ผ่านการ validate จาก wet lab test มาเรียบร้อยแล้วโดยมีค่าความไว (Sensitivity) อยู่ที่ 0.1% สำหรับการตรวจ mutation ในกลุ่ม wildtype โดยภายในชุด kit จะประกอบไปด้วยสีฟลูออเรสเซนต์ 2 ชนิดด้วยกันเพื่อตรวจวัด mutant และ wild-type sequences ใน 1 well           ชุด kit นี้จะอยู่ในรูปแบบหลอดที่พร้อมใช้งาน โดยมีความเข้มข้น 30x ประกอบไปด้วย probe-primer ของ wildtype และ mutate sequence ซึ่งผู้ใช้งานสามารถใช้ dPCR LNA Mutation Assays ร่วมกับ QIAcuity Probe PCR Kit ได้

     dPCR LNA Mutation Assays ได้มีการผสานนำ LNA Technology ซึ่งเป็น technology ในการเพิ่มความจำเพาะต่อ Nucleoside โดยมีการ “Lock” น้ำตาล Ribose ที่บริเวณ 2’-O Atom and The 4’-C Atom โดย Methylene Bridge ซึ่ง LNA™ Nucleosides มีองค์ประกอบทุกอย่างเหมือน DNA หรือ RNA อยู่แล้ว ดังนั้นจึงสามารถเข้าจับกับเบสคู่สมได้ตามหลัก Watson-Crick Base Pairing ซึ่ง Technology นี้ได้เพิ่มค่า Stability และ Specificity ของ Probe และ Primer มากยิ่งขึ้น.      ตัวอย่างแสดงการตรวจ BRAF mutation ที่ตำแหน่ง V600E (เป็นตำแหน่งที่เกิด mutation ของกรดอะมิโนที่ตำแหน่งที่ 600 ที่เกิดจากการเปลี่ยนแปลงของกรดอะมิโนจาก valine (V) เป็น glutamic acid (E)) โดยรูปที่ 2 แสดงการตรวจวัด BRAF V600E โดย 2D Scatter plot ซึ่งจากกราฟพบ 6 positive copies คิดเป็น 0.13% ในตัวอย่าง FFPE
